Principles of the Assay: This kit is an Enzyme-Linked Immunosorbent Assay (ELISA). The plate has been pre-coated with Fish FST/FS antibody. FST/FS present in the sample is added and binds to antibodies coated on the wells. And then biotinylated Fish FST/FS Antibody is added and binds to FST/FS in the sample. Then Streptavidin-HRP is added and binds to the Biotinylated FST/FS antibody. After incubation unbound Streptavidin-HRP is washed away during a washing step. Substrate solution is then added and color develops in proportion to the amount of Fish FST/FS. The reaction is terminated by addition of acidic stop solution and absorbance is measured at 450 nm.

Follistatin is a single-chain gonadal protein that specifically inhibits follicle-stimulating hormone release. The single FST gene encodes two isoforms, FST317 and FST344 containing 317 and 344 amino acids respectively, resulting from alternative splicing of the precursor mRNA. In a study in which 37 candidate genes were tested for linkage and association with polycystic ovary syndrome (PCOS) or hyperandrogenemia in 150 families, evidence was found for linkage between PCOS and follistatin.
