Principle of the Assay: FGF9 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-FGF9 antibody and an FGF9-HRP conjugate. The assay sample and buffer are incubated together with FGF9-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FGF9 concentration since FGF9 from samples and FGF9-HRP conjugate compete for the anti-FGF9 antibody binding site. Since the number of sites is limited, as more sites are occupied by FGF9 from the sample, fewer sites are left to bind FGF9-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FGF9 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes, including embryonic development, cell growth, morphogenesis, tissue repair, tumor growth and invasion. This protein was isolated as a secreted factor that exhibits a growth-stimulating effect on cultured glial cells. In nervous system, this protein is produced mainly by neurons and may be important for glial cell development. Expression of the mouse homolog of this gene was found to be dependent on Sonic hedgehog (Shh) signaling. Mice lacking the homolog gene displayed a male-to-female sex reversal phenotype, which suggested a role in testicular embryogenesis. [provided by RefSeq, Jul 2008]

Defects in FGF9 are the cause of multiple synostoses syndrome type 3 (SYNS3). Multiple synostoses syndrome is an autosomal dominant condition characterized by progressive joint fusions of the fingers, wrists, ankles and cervical spine, characteristic facies and progressive conductive deafness. Belongs to the heparin-binding growth factors family.
