Principle of the Assay: FGF19 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FGF19 antibody and an FGF19-HRP conjugate. The assay sample and buffer are incubated together with FGF19-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FGF19 concentration since FGF19 from samples and FGF19-HRP conjugate compete for the anti-FGF19 antibody binding site. Since the number of sites is limited, as more sites are occupied by FGF19 from the sample, fewer sites are left to bind FGF19-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FGF19 concentration in each sample is interpolated from this standard curve.

The protein encoded by this gene is a member of the fibroblast growth factor (FGF) family. FGF family members possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes including embryonic development cell growth, morphogenesis, tissue repair, tumor growth and invasion. This growth factor is a high affinity, heparin dependent ligand for FGFR4. Expression of this gene was detected only in fetal but not adult brain tissue. Synergistic interaction of the chick homolog and Wnt-8c has been shown to be required for initiation of inner ear development. [provided by RefSeq, Jul 2008]
