Specificity
This assay has high sensitivity and excellent specificity for detection of FBG. No significant cross-reactivity or interference between FBG and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between FBG and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This FBG ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of RatFBG. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: FBG ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FBG antibody and an FBG-HRP conjugate. The assay sample and buffer are incubated together with FBG-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FBG concentration since FBG from samples and FBG-HRP conjugate compete for the anti-FBG antibody binding site. Since the number of sites is limited, as more sites are occupied by FBG from the sample, fewer sites are left to bind FBG-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FBG concentration in each sample is interpolated from this standard curve.

Uniprot Description

Function: Fibrinogen has a double function: yielding monomers that polymerize into fibrin and acting as a cofactor in platelet aggregation.

Subunit structure: Heterohexamer; disulfide linked. Contains 2 sets of 3 non-identical chains (alpha, beta and gamma). The 2 heterotrimers are in head to head conformation with the N-termini in a small central domain

By similarity.

Subcellular location: Secreted.

Domain: A long coiled coil structure formed by 3 polypeptide chains connects the central nodule to the C-terminal domains (distal nodules). The long C-terminal ends of the alpha chains fold back, contributing a fourth strand to the coiled coil structure.

Post-translational modification: Conversion of fibrinogen to fibrin is triggered by thrombin, which cleaves fibrinopeptides A and B from alpha and beta chains, and thus exposes the N-terminal polymerization sites responsible for the formation of the soft clot. The soft clot is converted into the hard clot by factor XIIIA which catalyzes the epsilon-(gamma-glutamyl)lysine cross-linking between gamma chains (stronger) and between alpha chains (weaker) of different monomers.

Sequence similarities: Contains 1 fibrinogen C-terminal domain.

Product Notes

The Rat Fbg fgg (Catalog #AAA728433) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA728433 ELISA Kit recognizes Rat Fbg. It is sometimes possible for the material contained within the vial of "Fibrinogen, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
