Intended Uses: This FABP4 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine FABP4. This ELISA kit for research use only!

Principle of the Assay: FABP4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-FABP4 antibody and an FABP4-HRP conjugate. The assay sample and buffer are incubated together with FABP4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FABP4 concentration since FABP4 from samples and FABP4-HRP conjugate compete for the anti-FABP4 antibody binding site. Since the number of sites is limited, as more sites are occupied by FABP4 from the sample, fewer sites are left to bind FABP4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FABP4 concentration in each sample is interpolated from this standard curve.

NCBI Description

FABP4 encodes the fatty acid binding protein found in adipocytes. Fatty acid binding proteins are a family of small, highly conserved, cytoplasmic proteins that bind long-chain fatty acids and other hydrophobic ligands. It is thought that FABPs roles include fatty acid uptake, transport, and metabolism. [provided by RefSeq, Jul 2008]

Uniprot Description

FABP4: a lipid transport protein that is the predominant fatty acid binding protein found in adipocytes. Also expressed in macrophages. Binds both long chain fatty acids and retinoic acid. Delivers long-chain fatty acids and retinoic acid to their cognate receptors in the nucleus. Homodimer. Forms a beta-barrel structure that accommodates hydrophobic ligands in its interior. Interacts with PPARG. Monomer. FABP4 knockout mice fed a high-fat and high-calorie diet become obese but develop neither insulin resistance nor diabetes, suggesting that this protein might be a link between obesity and insulin resistance and diabetes. Mice deficient in both FABP4 and ApoE show protection against atherosclerosis when compared with mice deficient only in ApoE. Mice carrying a FABP4 genetic variant exhibit both reduced FABP4 expression and a reduced potential for developing type 2 diabetes and coronary heart disease. A related study in humans indicated a similar pattern, suggesting that FABP4 may be a potential therapeutic target in the treatment of these disorders. Belongs to the calycin superfamily,fatty-acid binding protein (FABP) family.

Product Notes

The Bovine FABP4 fabp4 (Catalog #AAA736377) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA736377 ELISA Kit recognizes Bovine FABP4. It is sometimes possible for the material contained within the vial of "Fatty Acid Binding Protein 4, Adipocyte, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
