Intended Uses: This F11 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Monkey F11. This ELISA kit for research use only!

Principle of the Assay: F11 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-F11 antibody and an F11-HRP conjugate. The assay sample and buffer are incubated together with F11-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the F11 concentration since F11 from samples and F11-HRP conjugate compete for the anti-F11 antibody binding site. Since the number of sites is limited, as more sites are occupied by F11 from the sample, fewer sites are left to bind F11-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The F11 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes coagulation factor XI of the blood coagulation cascade. This protein is present in plasma as a zymogen, which is a unique plasma coagulation enzyme because it exists as a homodimer consisting of two identical polypeptide chains linked by disulfide bonds. During activation of the plasma factor XI, an internal peptide bond is cleaved by factor XIIa (or XII) in each of the two chains, resulting in activated factor XIa, a serine protease composed of two heavy and two light chains held together by disulfide bonds. This activated plasma factor XI triggers the middle phase of the intrisic pathway of blood coagulation by activating factor IX. Defects in this factor lead to Rosenthal syndrome, a blood coagulation abnormality. [provided by RefSeq, Jul 2008]

Defects in F11 are the cause of factor XI deficiency (FA11D); also known as plasma thromboplastin antecedent deficiency or Rosenthal syndrome. It is a hemorrhagic disease characterized by reduced levels and activity of factor XI resulting in moderate bleeding symptoms, usually occurring after trauma or surgery. Patients usually do not present spontaneous bleeding but women can present with menorrhagia. Hemorrhages are usually moderate. Belongs to the peptidase S1 family. Plasma kallikrein subfamily. 2 isoforms of the human protein are produced by alternative splicing.
