Intended Uses: This EP3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Chicken EP3. This ELISA kit for research use only!

Principle of the Assay: EP3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-EP3 antibody and an EP3-HRP conjugate. The assay sample and buffer are incubated together with EP3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the EP3 concentration since EP3 from samples and EP3-HRP conjugate compete for the anti-EP3 antibody binding site. Since the number of sites is limited, as more sites are occupied by EP3 from the sample, fewer sites are left to bind EP3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The EP3 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the G-protein coupled receptor family. This protein is one of four receptors identified for prostaglandin E2 (PGE2). This receptor may have many biological functions, which involve digestion, nervous system, kidney reabsorption, and uterine contraction activities. Studies of the mouse counterpart suggest that this receptor may also mediate adrenocorticotropic hormone response as well as fever generation in response to exogenous and endogenous stimuli. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Aug 2009]

Uniprot Description

PTGER3: Receptor for prostaglandin E2 (PGE2); the EP3 receptor may be involved in inhibition of gastric acid secretion, modulation of neurotransmitter release in central and peripheral neurons, inhibition of sodium and water reabsorption in kidney tubulus and contraction in uterine smooth muscle. The activity of this receptor can couple to both the inhibition of adenylate cyclase mediated by G-I proteins, and to an elevation of intracellular calcium. The various isoforms have identical ligand binding properties but can interact with different second messenger systems. Belongs to the G-protein coupled receptor 1 family. 7 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, multi-pass; Membrane protein, integral; Receptor, GPCR; GPCR, family 1

Chromosomal Location of Human Ortholog: 1p31.2

Cellular Component: integral to plasma membrane; plasma membrane; integral to membrane; nuclear envelope

Molecular Function: ligand-dependent nuclear receptor activity; prostaglandin E receptor activity

Biological Process: G-protein signaling, coupled to cAMP nucleotide second messenger; cell death; G-protein coupled receptor protein signaling pathway; elevation of cytosolic calcium ion concentration; intracellular receptor-mediated signaling pathway; transcription, DNA-dependent; bicarbonate transport; positive regulation of fever; response to lipopolysaccharide; G-protein signaling, coupled to IP3 second messenger (phospholipase C activating); fever

Product Notes

The Chicken EP3 ptger3 (Catalog #AAA746802) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA746802 ELISA Kit recognizes Chicken EP3. It is sometimes possible for the material contained within the vial of "Prostaglandin E Receptor 3, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
