Principle of the Assay: The design of this assay is based on a sandwich Enzyme-Linked Immunosorbent Assay (ELISA). The microtiter plate provided in this kit has been pre-coated with a monoclonal antibody specific to Human LT-alpha (TNF-beta). Samples are pipetted into these wells. Nonbound Human LT-alpha (TNF-beta) and other components of the sample should be removed by washing, then monoclonal antibody specific to Human LT-alpha (TNF-beta) added. In order to quantitatively determine the amount of Human LT-alpha (TNF-beta) present in the sample, Streptavidin Horseradish Peroxidase (HRP) should be added to each microplate well. The final step, a TMB-substrate solution added to each well. Finally, a sulfuric acid solution is added and the resulting yellow colored product is measured at 450nm. Since the increases in absorbency is directly proportional to the amount of captured Human LT-alpha (TNF-beta)!!Background/Introduction: Lymphotoxin-alpha (LT-alpha) is a protein that in humans is encoded by the LTA gene. Lymphotoxin alpha, a member of the tumor necrosis factor family, is a cytokine produced by lymphocytes. LTA is highly inducible, secreted, and exists as homotrimeric molecule. LTA forms heterotrimers with lymphotoxin-beta, which anchors lymphotoxin-alpha to the cell surface. LTA mediates a large variety of inflammatory, immunostimulatory, and antiviral responses. LTA is also involved in the formation of secondary lymphoid organs during development and plays a role in apoptosis. In LT-alpha knockout mice, all Peyer's patches and lymph nodes will fail to develop indicating LT-alpha's importance in immunological development. Lymphotoxin alpha has been shown to interact with LTB.
