Anti-PM-Scl ELISA
Principle of the Assay: Anti Alpha Fodrin IgG/IgA ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-Anti Alpha Fodrin IgG/IgA antibody and an Anti Alpha Fodrin IgG/IgA-HRP conjugate. The assay sample and buffer are incubated together with Anti Alpha Fodrin IgG/IgA-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the Anti Alpha Fodrin IgG/IgA concentration since Anti Alpha Fodrin IgG/IgA from samples and Anti Alpha Fodrin IgG/IgA-HRP conjugate compete for the anti-Anti Alpha Fodrin IgG/IgA antibody binding site. Since the number of sites is limited, as more sites are occupied by Anti Alpha Fodrin IgG/IgA from the sample, fewer sites are left to bind Anti Alpha Fodrin IgG/IgA-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The Anti Alpha Fodrin IgG/IgA concentration in each sample is interpolated from this standard curve.

Putative catalytic component of the RNA exosome complex which has 3'->5' exoribonuclease activity and participates in a multitude of cellular RNA processing and degradation events. In the nucleus, the RNA exosome complex is involved in proper maturation of stable RNA species such as rRNA, snRNA and snoRNA, in the elimination of RNA processing by-products and non-coding 'pervasive' transcripts, such as antisense RNA species and promoter-upstream transcripts (PROMPTs), and of mRNAs with processing defects, thereby limiting or excluding their export to the cytoplasm. The RNA exosome may be involved in Ig class switch recombination (CSR) and/or Ig variable region somatic hypermutation (SHM) by targeting AICDA deamination activity to transcribed dsDNA substrates. In the cytoplasm, the RNA exosome complex is involved in general mRNA turnover and specifically degrades inherently unstable mRNAs containing AU-rich elements (AREs) within their 3' untranslated regions, and in RNA surveillance pathways, preventing translation of aberrant mRNAs. It seems to be involved in degradation of histone mRNA. EXOSC10 has 3'-5' exonuclease activity (). EXOSC10 is required for nucleolar localization of C1D and probably mediates the association of SKIV2L2, C1D and MPP6 wth the RNA exosome involved in the maturation of 5.8S rRNA.
