Intended Uses: The rat IL-18 ELISA has been designed for the quantitative detection of IL-18 concentrations in rat serum, plasma, body fluids, tissue lysates or cell culture supernates. For research use only,

Principle of the Assay: This kit is based on a sandwich enzyme-linked immune-sorbent assay technology. Anti-IL-18 polyclonal antibody is precoated onto the 96 well plate. The biotin conjugated anti-IL-18 polyclonal antibody is used as the detection antibody. The standards, test samples and biotin conjugated detection antibody are added to the wells subsequently and washed with wash buffer. Avidin-Biotin-Peroxidase Conjugate is added, and unbound conjugate is washed away with wash buffer. TMB substrate is added to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The optical density of the yellow color is proportional to the analyte captured in plate. Read the O.D. absorbance at 450nm in a microtiter plate reader, and calculate the concentration of the analyte in the sample by taking into consideration the dilution factor of the sample.

Background/Introduction: Interleukin-18 (IL18), also known as interferon-gamma inducing factor, in humans is encoded by the IL18 gene, this gene contains 6 exons, and maps to 11q22.2-q22.3. It is a cytokine that belongs to the IL-1 superfamily and is produced by macrophages and other cells. It works by binding to the interleukin-18 receptor, and together with IL-12 it induces cellmediated immunity following infection with microbial products like lipopolysaccharide (LPS). IL-18 has been implicated as an inflammatory mediator of Hashimoto's thyroiditis, the most common cause of autoimmune hypothyroidism. IL-18 is also able to induce severe inflammatory reactions, which suggests its role in certain inflammatory disorders.

Product Notes

The IL-18 il18 (Catalog #AAA495380) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "IL-18, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
