Related Product Information for eIF3A elisa kit
Principle of the assay: EIF-3A ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-EIF-3A antibody and an EIF-3A-HRP conjugate. The assay sample and buffer are incubated together with EIF-3A-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the EIF-3A concentration since EIF-3A from samples and EIF-3A-HRP conjugate compete for the anti-EIF-3A antibody binding site. Since the number of sites is limited, as more sites are occupied by EIF-3A from the sample, fewer sites are left to bind EIF-3A-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The EIF-3A concentration in each sample is interpolated from this standard curve.

Uniprot Description

eIF3-theta: Component of the eukaryotic translation initiation factor 3 (eIF-3) complex, which is required for several steps in the initiation of protein synthesis. The eIF-3 complex associates with the 40S ribosome and facilitates the recruitment of eIF-1, eIF-1A, eIF-2:GTP:methionyl-tRNAi and eIF-5 to form the 43S preinitiation complex (43S PIC). The eIF-3 complex stimulates mRNA recruitment to the 43S PIC and scanning of the mRNA for AUG recognition. The eIF-3 complex is also required for disassembly and recycling of posttermination ribosomal complexes and subsequently prevents premature joining of the 40S and 60S ribosomal subunits prior to initiation. Interacts with EIF4G1. Component of the eukaryotic translation initiation factor 3 (eIF-3) complex, which is composed of 13 subunits: EIF3A, EIF3B, EIF3C, EIF3D, EIF3E, EIF3F, EIF3G, EIF3H, EIF3I, EIF3J, EIF3K, EIF3L and EIF3M. The eIF-3 complex appears to include 3 stable modules: module A is composed of EIF3A, EIF3B, EIF3G and EIF3I; module B is composed of EIF3F, EIF3H, and EIF3M; and module C is composed of EIF3C, EIF3D, EIF3E, EIF3L and EIF3K. EIF3C of module C binds EIF3B of module A and EIF3H of module B, thereby linking the three modules. EIF3J is a labile subunit that binds to the eIF-3 complex via EIF3B. The eIF-3 complex interacts with RPS6KB1 under conditions of nutrient depletion. Mitogenic stimulation leads to binding and activation of a complex composed of MTOR and RPTOR, leading to phosphorylation and release of RPS6KB1 and binding of EIF4B to eIF-3. Also interacts with KRT7 and PIWIL2. Belongs to the eIF-3 subunit A family.

Product Notes

The Human eIF3A eif3a (Catalog #AAA720629) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA720629 ELISA Kit recognizes Human eIF3A. It is sometimes possible for the material contained within the vial of "Eukaryotic Translation Initiation Factor 3A, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
