Reconstitution
Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/m. Do not vortex. This solution can be stored at 2-8°C. For extended storage, itis recommended to further dilute in buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
Preparation and Storage
The lyophilized protein is stable at 4°C. Reconstituted working aliquots are stable at 2°C to 8°C and -20°C to -80°C.

GDNF is a disulfide-linked homodimeric neurotrophic factor structurally related to Artemin, Neurturin and Persephin. These proteins belong to the cysteine-knot superfamily of growth factors that assume stable dimeric protein structures. GDNF signals through a multicomponent receptor system, composed of a RET and one of the four GFRalpha (alpha1-alpha4) receptors. GDNF specifically promotes dopamine uptake and survival and morphological differentiation of midbrain neurons. Using Parkinson\'s disease mouse model, GDNF has been shown to improve conditions such as bradykinesia, rigidity, and postural instability. The functional human GDNF ligand is a disulfide-linked homodimer, of two 15 kDa polypeptide chains called monomers. Each monomer contains seven conserved cysteine residues, one of which (Cys 101) is used for inter-chain disulfide bridging and the others are involved in intramolecular ring formation known as the cysteine knot configuration.

Uniprot Description

Function: Neurotrophic factor that enhances survival and morphological differentiation of dopaminergic neurons and increases their high-affinity dopamine uptake.

Subunit structure: Homodimer; disulfide-linked.

Subcellular location: Secreted.

Tissue specificity: Expressed in both the central nervous system (CNS) and in non-CNS tissues. Expressed in a highly dynamic pattern in the anterior neuroectoderm during the early stages of neurogenesis between E7.5 and E10.5. Beginning at E10.5, expression begins in mesenchymal tissues of several organs including the digestive tract, kidney, testis, frontonasal mass, tooth primordium, tongue, mandible, whisker follicles, ear, eye, limb bud and in distinct regions of the brain. Also expressed in the heart, ileum, liver and muscle. Ref.2 Ref.4

Developmental stage: Expressed begins by E7.5, reaches its peak around E9.5 and declines considerably after E10.5. Ref.2

Induction: Expression in C6 glioma cells was transiently induced by treatment with phorbol myristate acetate (PMA), but not by forskolin. Ref.3

Sequence similarities: Belongs to the TGF-beta family. GDNF subfamily.
