The recombinant human IL1beta consisting of 154 amino acids migrates with an apparent molecular mass of 17 kDa as estimated in SDS-PAGE under reducing conditions.
Activity
1. Measured by its binding ability in a functional ELISA. Immobilized recombinant human IL1B at 2 mug/mL (100mul/well) can bind recombinant human IL1R2 with a linear range of 6.25-400 ng/ml. 2. Measured in a cell proliferation assay using D10.G4.1 mouse helper T cells. The ED50 for this effect is typically 2-8 pg/mL. 3. Measured by its binding ability in a functional ELISA. 4. Immobilized human IL1B at 10 mug/mL (100 mul/well) can bind human IL1R1, The EC50 of human IL1R1 is 0.48 mug/mL. 5. Measured by its ability to induce Interferon gamma secretion by human natural killer lymphoma NK-92 cells. The ED50 for this effect is 0.4-2 ng/mL.
Usage Notes
Always centrifuge tubes before opening. It is recommended to reconstitute the lyophilized Human IL-1 beta protein in sterile ddH2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions.
Preparation and Storage
Item should be stored desiccated below -20 degree C. Upon reconstitution, the protein should be stored at 4 degree C between 2-7 days and for future use below -20 degree C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.

Interleukin-1 beta (IL1 beta or IL1B) also known as catabolin, is a member of the interleukin 1 cytokine family. IL1 is a pleiotropic cytokine. It is involved in the inflammatory response, cell growth, and tissue repair in the cortex. The IL1 superfamily consists of three members, IL1A (IL1 alpha), IL1B (IL1 beta), and IL1 receptor antagonist (IL1Ra). In clinical, it has been reported that Interleukin (IL)-1 may influence Th1/ Th2 immune responsiveness and has been implicated in the establishment of successful pregnancy. Proinflammatory interleukin (IL)-1 gene polymorphisms associated with high levels of IL-1beta activity increase the risk for hypochlorhydria and distal gastric carcinoma. IL1B polymorphisms may be involved in susceptibility to SSc. Moreover, the IL2-384-G allele may be a marker for the limited phenotype of systemic sclerosis (SSc).

NCBI Description

The protein encoded by this gene is a member of the interleukin 1 cytokine family. This cytokine is produced by activated macrophages as a proprotein, which is proteolytically processed to its active form by caspase 1 (CASP1/ICE). This cytokine is an important mediator of the inflammatory response, and is involved in a variety of cellular activities, including cell proliferation, differentiation, and apoptosis. The induction of cyclooxygenase-2 (PTGS2/COX2) by this cytokine in the central nervous system (CNS) is found to contribute to inflammatory pain hypersensitivity. This gene and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2. [provided by RefSeq, Jul 2008]
