Solubility
It is recommended to reconstitute the lyophilized Recombinant TPO in sterile 18Momega-cm H2O not less than 100ug/ml, which can then be further diluted to other aqueous solutions.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm.
2. Analysis by RP-HPLC, using a calibrated solution of TPO as a Reference Standard.

Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
Endotoxin
Less than 0.1 ng/ug (IEU/ug) of TPO.
Formulation
Lyophilized from a concentrated (1mg/ml) solution in water containing no additives.
Biological Activity
Recombinant TPO is fully biologically active when compared to standard. The ED50 as determined by the dose-dependant stimulation of MO7e cells is less then 1 ng/ml, corresponding to a Specific Activity of 1 x 10 6 IU/mg.
Preparation and Storage
Lyophilized Recombinant TPO although stable at room temperature for 3 weeks, should be stored desiccated below -18 degree C. Upon reconstitution Recombinant TPO should be stored at 4 degree C between 2-7 days and for future use below -18 degree C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please avoid freeze-thaw cycles.

NCBI Description

Megakaryocytopoiesis is the cellular development process that leads to platelet production. The main functional protein encoded by this gene is a humoral growth factor that is necessary for megakaryocyte proliferation and maturation, as well as for thrombopoiesis. This protein is the ligand for MLP/C_MPL, the product of myeloproliferative leukemia virus oncogene. Mutations in this gene are the cause of thrombocythemia 1. Alternative promoter usage and differential splicing result in multiple transcript variants differing in the 5' UTR and/or coding region. Multiple AUG codons upstream of the main open reading frame (ORF) have been identified, and these upstream AUGs inhibit translation of the main ORF at different extent. THPO: Lineage-specific cytokine affecting the proliferation and maturation of megakaryocytes from their committed progenitor cells. It acts at a late stage of megakaryocyte development. It may be the major physiological regulator of circulating platelets. Defects in THPO are the cause of thrombocythemia type 1 (THCYT1). A myeloproliferative disorder characterized by elevated platelet levels due to sustained proliferation of megakaryocytes, and frequently lead to thrombotic and haemorrhagic complications. Belongs to the EPO/TPO family. 3 isoforms of the human protein are produced by alternative splicing.
