Activity

(Vimentin (VIM) is a type III intermediate filament (IF) protein that is expressed in mesenchymal cells.vimentin is the major cytoskeletal component of mesenchymal cells. Because of this, vimentin is often used as a marker of mesenchymally-derived cells or cells undergoing an epithelial-to-mesenchymal transition (EMT) during both normal development and metastatic progression. Besides, Keratin 20 (KRT20) has been identified as an interactor of VIM, thus a binding ELISA assay was conducted to detect the interaction of recombinant human VIM and recombinant human KRT20. Briefly, VIM were diluted serially in PBS, with 0.01% BSA (pH 7.4). Duplicate samples of 100?l then transferred to KRT20-coated microtiter wells and incubated for 2h at 37?. Wells were washed with PBST and incubated for 1h with anti-VIM pAb, then aspirated and washed 3 times. After incubation with HRP labelled secondary antibody, wells were aspirated and washed 3 times. With the addition of substrate solution, wells were incubated 15-25 minutes at 37?. Finally, add 50µl stop solution to the wells and read at 450nm immediately. The binding activity of VIM and KRT20 was shown in Figure 1, and this effect was in a dose dependent manner.)

NCBI Description

This gene encodes a member of the intermediate filament family. Intermediate filamentents, along with microtubules and actin microfilaments, make up the cytoskeleton. The protein encoded by this gene is responsible for maintaining cell shape, integrity of the cytoplasm, and stabilizing cytoskeletal interactions. It is also involved in the immune response, and controls the transport of low-density lipoprotein (LDL)-derived cholesterol from a lysosome to the site of esterification. It functions as an organizer of a number of critical proteins involved in attachment, migration, and cell signaling. Mutations in this gene causes a dominant, pulverulent cataract.[provided by RefSeq, Jun 2009]

Product Notes

The VIM vim (Catalog #AAA2125335) is an Active Protein produced from E Coli and is intended for research purposes only. The product is available for immediate purchase. AAA Biotech's Vimentin (VIM) can be used in a range of immunoassay formats including, but not limited to, Cell culture, Activity Assays. Researchers should empirically determine the suitability of the VIM vim for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. The amino acid sequence is listed below: Ser2~Glu46 6. It is sometimes possible for the material contained within the vial of "Vimentin (VIM), Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
