Biological Activity
Recombinant Human I-TAC is fully biologically active when compared to standard. The Activity is calculated by the ability to chemoattract IL-2 activated T-cells using a concentration of 0.1-10 ng/ml.
Dimers and Aggregates
Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
Endotoxin Level
0.1ng/ug (IEU/ug) of Human I-TAC.
Protein Content
Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm.
2. Analysis by RP-HPLC, using a calibrated solution of Human I-TAC as a Reference Standard.
Solubility
It is recommended to reconstitute the lyophilized Recombinant Human I-TAC in sterile 18M -cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
Preparation and Storage
-20 degree C
Stability: Lyophilized I-TAC although stable at room temperature for 3 weeks, should be stored desiccated below -180C. Upon reconstitution I-TAC should be stored at 40C between 2-7 days and for future use below -180C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please avoid freeze-thaw cycles.

CXCL11 (ITAC) is one of three chemokines known to bind the receptor CXCR3, the two others being CXCL9 (Mig) and CXCL10 (IP-10). CXCL11 differs from the other CXCR3 ligands in both the strength and the particularities of its receptor interactions: It has a higher affinity, is a stronger agonist, and behaves differently when critical N-terminal residues are deleted CXCL11 takes on the canonical chemokine fold but exhibits greater conformational flexibility than has been observed for related chemokines under the same sample conditions. Unlike related chemokines such as IP-10 and IL-8, ITAC does not appear to form dimers at millimolar concentrations. The origin for this behavior can be found in the solution structure, which indicates a beta-bulge in beta-strand 1 that distorts the dimerization interface used by other CXC chemokines.

Recombinant Human I-TAC (Interferon-inducible T-cell alpha chemoattractant) produced in E.Coli is a single, non-glycosylated, polypeptide chain containing 73 amino acids and having a molecular mass of 8300 Dalton. Recombinant Human I-TAC is purified by proprietary chromatographic techniques.

Chemokines are a group of small (approximately 8 to 14 kD), mostly basic, structurally related molecules that regulate cell trafficking of various types of leukocytes through interactions with a subset of 7-transmembrane, G protein-coupled receptors. Chemokines also play fundamental roles in the development, homeostasis, and function of the immune system, and they have effects on cells of the central nervous system as well as on endothelial cells involved in angiogenesis or angiostasis. Chemokines are divided into 2 major subfamilies, CXC and CC. This gene is a CXC member of the chemokine superfamily. Its encoded protein induces a chemotactic response in activated T-cells and is the dominant ligand for CXC receptor-3. The gene encoding this protein contains 4 exons and at least three polyadenylation signals which might reflect cell-specific regulation of expression. IFN-gamma is a potent inducer of transcription of this gene. [provided by RefSeq]

Uniprot Description

Function: Chemotactic for interleukin-activated T-cells but not unstimulated T-cells, neutrophils or monocytes. Induces calcium release in activated T-cells. Binds to CXCR3. May play an important role in CNS diseases which involve T-cell recruitment. May play a role in skin immune responses.

Subcellular location: Secreted.

Tissue specificity: High levels in peripheral blood leukocytes, pancreas and liver astrocytes. Moderate levels in thymus, spleen and lung. Low levels in placenta, prostate and small intestine. Also found in epidermal basal layer keratinocytes in skin disorders.

Induction: By IFNG/IFN-gamma and IFNB1/IFN-beta. Induction by IFNG/IFN-gamma is enhanced by TNF in monocytes, dermal fibroblasts and endothelial cells, and by IL1/interleukin-1 in astrocytes.

Sequence similarities: Belongs to the intercrine alpha (chemokine CxC) family.

Mass spectrometry: Molecular mass is 8303 Da from positions 22 - 94. Determined by MALDI. Ref.4
