Preparation and Storage

Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti-human DCNantibody was pre-coated onto 96-well plates. And the biotin conjugated anti-human DCN antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human DCN amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration ofhuman DCNcan be calculated.

NCBI Description

This gene encodes a member of the small leucine-rich proteoglycan family of proteins. Alternative splicing results in multiple transcript variants, at least one of which encodes a preproprotein that is proteolytically processed to generate the mature protein. This protein plays a role in collagen fibril assembly. Binding of this protein to multiple cell surface receptors mediates its role in tumor suppression, including a stimulatory effect on autophagy and inflammation and an inhibitory effect on angiogenesis and tumorigenesis. This gene and the related gene biglycan are thought to be the result of a gene duplication. Mutations in this gene are associated with congenital stromal corneal dystrophy in human patients. Corneal dystrophies are inherited, bilateral, primary alterations of the cornea that are not associated with prior inflammation or secondary to systemic disease. Most show autosomal dominant inheritance. Belongs to the small leucine-rich proteoglycan (SLRP) family. SLRP class I subfamily. 5 isoforms of the human protein are produced by alternative splicing.
