Principle of the assay: CX31 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CX31 antibody and an CX31-HRP conjugate. The assay sample and buffer are incubated together with CX31-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CX31 concentration since CX31 from samples and CX31-HRP conjugate compete for the anti-CX31 antibody binding site. Since the number of sites is limited, as more sites are occupied by CX31 from the sample, fewer sites are left to bind CX31-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CX31 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene is a member of the connexin gene family. The encoded protein is a component of gap junctions, which are composed of arrays of intercellular channels that provide a route for the diffusion of low molecular weight materials from cell to cell. Mutations in this gene can cause non-syndromic deafness or erythrokeratodermia variabilis, a skin disorder. Alternative splicing results in multiple transcript variants encoding the same protein. [provided by RefSeq, Jul 2008]

Uniprot Description

GJB3: One gap junction consists of a cluster of closely packed pairs of transmembrane channels, the connexons, through which materials of low MW diffuse from one cell to a neighboring cell. Defects in GJB3 are a cause of erythrokeratodermia variabilis (EKV). EKV is a genodermatosis characterized by the appearance of two independent skin lesions: transient figurate erythematous patches and hyperkeratosis that is usually localized but occasionally occurs in its generalized form. Clinical presentation varies significantly within a family and from one family to another. Palmoplantar keratoderma is present in around 50% of cases. Defects in GJB3 are the cause of deafness autosomal dominant type 2B (DFNA2B). DFNA2 is a form of sensorineural hearing loss. Sensorineural deafness results from damage to the neural receptors of the inner ear, the nerve pathways to the brain, or the area of the brain that receives sound information. Belongs to the connexin family. Beta-type (group I) subfamily.

Product Notes

The Rabbit CX31 gjb3 (Catalog #AAA720473) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA720473 ELISA Kit recognizes Rabbit CX31. It is sometimes possible for the material contained within the vial of "Connexin 31, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
