Principle of the Assay: CVL ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CVL antibody and an CVL-HRP conjugate. The assay sample and buffer are incubated together with CVL-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CVL concentration since CVL from samples and CVL-HRP conjugate compete for the anti-CVL antibody binding site. Since the number of sites is limited, as more sites are occupied by CVL from the sample, fewer sites are left to bind CVL-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CVL concentration in each sample is interpolated from this standard curve.

NCBI Description

The cytoplasmic peripheral membrane protein encoded by this gene functions as a protein-tyrosine kinase substrate in microvilli. As a member of the ERM protein family, this protein serves as an intermediate between the plasma membrane and the actin cytoskeleton. This protein plays a key role in cell surface structure adhesion, migration and organization, and it has been implicated in various human cancers. A pseudogene located on chromosome 3 has been identified for this gene. Alternatively spliced variants have also been described for this gene. [provided by RefSeq, Jul 2008]
