Canine Cardiac Troponin 1 ELISA Kit
Principle of the Assay: cTnI ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-cTnI antibody and an cTnI-HRP conjugate. The assay sample and buffer are incubated together with cTnI-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the cTnI concentration since cTnI from samples and cTnI-HRP conjugate compete for the anti-cTnI antibody binding site. Since the number of sites is limited, as more sites are occupied by cTnI from the sample, fewer sites are left to bind cTnI-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The cTnI concentration in each sample is interpolated from this standard curve.

Troponin I (TnI), along with troponin T (TnT) and troponin C (TnC), is one of 3 subunits that form the troponin complex of the thin filaments of striated muscle. TnI is the inhibitory subunit; blocking actin-myosin interactions and thereby mediating striated muscle relaxation. The TnI subfamily contains three genes: TnI-skeletal-fast-twitch, TnI-skeletal-slow-twitch, and TnI-cardiac. This gene encodes the TnI-cardiac protein and is exclusively expressed in cardiac muscle tissues. Mutations in this gene cause familial hypertrophic cardiomyopathy type 7 (CMH7) and familial restrictive cardiomyopathy (RCM). [provided by RefSeq, Jul 2008]
