Sandwich
Detection Range
0.312 ng/ml -20 ng/ml.
Sensitivity
The minimum detectable dose of human CLEC9A is typically less than 0.078 ng/ml.The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
Intra-assay Precision
Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess.
Inter-assay Precision
Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
Preparation and Storage
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CLEC9A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CLEC9A present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CLEC9A is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CLEC9A bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

CLEC9A is a group V C-type lectin-like receptor (CTLR) that functions as an activation receptor and is expressed on myeloid lineage cells (Huysamen et al., 2008 [PubMed 18408006]).[supplied by OMIM, Aug 2008]

Uniprot Description

CLEC9A: Functions as an endocytic receptor on a small subset of myeloid cells and induces intracellular signaling via spleen tyrosine kinase (Syk). Acts as a receptor for necrotic cells and can mediate the cross-presentation of dead-cell associated antigens in a Syk-dependent manner.

Protein type: Membrane protein, integral; Receptor, misc.

Chromosomal Location of Human Ortholog: 12p13.2

Cellular Component: cell surface; integral to membrane

Molecular Function: carbohydrate binding

Biological Process: receptor-mediated endocytosis; positive regulation of cytokine secretion

Product Notes

The Human CLEC9A clec9a (Catalog #AAA922282) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA922282 ELISA Kit recognizes Human CLEC9A. It is sometimes possible for the material contained within the vial of "C-type lectin domain family 9, member A, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
