Intended Uses: For the quantitative detection of Human Cytokeratin 18 (CK-18) concentration in serum, plasma, cell culture supernates, cell lysates and other biological fluids.

Principle of the Assay: This assay employs the sandwich enzyme immunoassay technique.The microtiter plate provided in this kit has been pre-coated with an antibody specific to CK-18. Standards or samples are then added to the appropriate microtiter plate wells with a Horseradish Peroxidase (HRP)-conjugated antibody preparation specific for CK-18 and incubated. Then substrate solution A and B are added to each well. Only those wells that contain CK-18, HRP-conjugated antibody will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of CK-18 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

NCBI Description

KRT18 encodes the type I intermediate filament chain keratin 18. Keratin 18, together with its filament partner keratin 8, are perhaps the most commonly found members of the intermediate filament gene family. They are expressed in single layer epithelial tissues of the body. Mutations in this gene have been linked to cryptogenic cirrhosis. Two transcript variants encoding the same protein have been found for this gene. [provided by RefSeq, Jul 2008]

Defects are a cause of cryptogenic cirrhosis.
