Molecular Structure
A soluble molecule consisting of the extracellular domain (132 aa) of human CD134L fused to the extracellular domain (167aa) of murine CD8 alpha.

Performance
Five x 105 cultured human HPB-MLT cells per tube were washed and incubated 45 minutes on ice with 80 ul of CD252-muCD8 at a concentration of 5 ug/ml. Cells were washed twice and incubated with 2 degree reagent anti-mouse CD8a/R-PE, after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.28 log10 fluorescent units. Binding was blocked when the reagent was pre incubated with a 20-fold excess of CD134-muIg.
Production
Fusion protein from (low FBS containing) tissue culture supernatant of transfectants was purified using affinity chromatography. Product was 0.2 um filtered and vialed under aseptic conditions.
Buffer
50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl.
Preparation and Storage
Store at 2 - 5 degree C. Open under aseptic conditions. Freeze/Thawing is not recommended.
Product should retain activity for at least 6 months after shipping date when stored as recommended.

Information: Human CD134L (OX-40 Ligand) is a type II membrane protein with homology to TNF which is expressed on activated B cells and activated endothelial cells. CD134L binds to CD134 present on activated T cells, providing a costimulatory signal (1). This pathway seems to be more effective for costimulation of CD4+ helper T cells than for CD8+ effector T cells (4). Blockade of this interaction in mouse abrogated immunological effects in several models of immflamation and rejection (5,6,7,8). CD134L-muCD8 fusion protein binds to CD134 positive cells in Flow cytometry.
