Intended Uses: This CD40L ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Rat CD40L. This ELISA kit for research use only!

Principle of the Assay: CD40L ELISA kit applies the competitive enzyme immunoassay technique utilizing a Polyclonal anti-CD40L antibody and an CD40L-HRP conjugate. The assay sample and buffer are incubated together with CD40L-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CD40L concentration since CD40L from samples and CD40L-HRP conjugate compete for the anti-CD40L antibody binding site. Since the number of sites is limited, as more sites are occupied by CD40L from the sample, fewer sites are left to bind CD40L-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CD40L concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is expressed on the surface of T cells. It regulates B cell function by engaging CD40 on the B cell surface. A defect in this gene results in an inability to undergo immunoglobulin class switch and is associated with hyper-IgM syndrome. [provided by RefSeq]

Uniprot Description

Function: Mediates B-cell proliferation in the absence of co-stimulus as well as IgE production in the presence of IL-4. Involved in immunoglobulin class switching. Ref.10Release of soluble CD40L from platelets is partially regulated by GP IIb/IIIa, actin polymerization, and an matrix metalloproteinases (MMP) inhibitor-sensitive pathway. Ref.10

Subunit structure: Homotrimer.

Subcellular location: Cell membrane; Single-pass type II membrane protein. CD40 ligand, soluble form: Secreted.

Tissue specificity: Specifically expressed on activated CD4+ T-lymphocytes.

Post-translational modification: The soluble form derives from the membrane form by proteolytic processing.N-linked glycan is a mixture of high mannose and complex type. Glycan structure does not influence binding affinity to CD40.Not O-glycosylated. Ref.9

Involvement in disease: Defects in CD40LG are the cause of X-linked immunodeficiency with hyper-IgM type 1 (HIGM1) [

MIM:308230]; also known as X-linked hyper IgM syndrome (XHIM). HIGM1 is an immunoglobulin isotype switch defect characterized by elevated concentrations of serum IgM and decreased amounts of all other isotypes. Affected males present at an early age (usually within the first year of life) recurrent bacterial and opportunistic infections, including Pneumocystis carinii pneumonia and intractable diarrhea due to cryptosporidium infection. Despite substitution treatment with intravenous immunoglobulin, the overall prognosis is rather poor, with a death rate of about 10% before adolescence. Ref.3 Ref.13 Ref.14 Ref.15 Ref.16 Ref.17 Ref.18 Ref.19 Ref.20 Ref.21

Sequence similarities: Belongs to the tumor necrosis factor family.

Product Notes

The Rat CD40L cd40lg (Catalog #AAA721043) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA721043 ELISA Kit recognizes Rat CD40L. It is sometimes possible for the material contained within the vial of "Cluster of differentiation 40 ligand, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
