Rat Chemokine C-C motif ligand 2 ELISA Kit
Principle of the Assay: CCL2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CCL2 antibody and an CCL2-HRP conjugate. The assay sample and buffer are incubated together with CCL2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CCL2 concentration since CCL2 from samples and CCL2-HRP conjugate compete for the anti-CCL2 antibody binding site. Since the number of sites is limited, as more sites are occupied by CCL2 from the sample, fewer sites are left to bind CCL2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CCL2 concentration in each sample is interpolated from this standard curve.

This gene is one of several cytokine genes clustered on the q-arm of chromosome 17. Chemokines are a superfamily of secreted proteins involved in immunoregulatory and inflammatory processes. The superfamily is divided into four subfamilies based on the arrangement of N-terminal cysteine residues of the mature peptide. This chemokine is a member of the CC subfamily which is characterized by two adjacent cysteine residues. This cytokine displays chemotactic activity for monocytes and basophils but not for neutrophils or eosinophils. It has been implicated in the pathogenesis of diseases characterized by monocytic infiltrates, like psoriasis, rheumatoid arthritis and atherosclerosis. It binds to chemokine receptors CCR2 and CCR4.
