Principle of the Assay: CASP4 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-CASP4 antibody and an CASP4-HRP conjugate. The assay sample and buffer are incubated together with CASP4-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CASP4 concentration since CASP4 from samples and CASP4-HRP conjugate compete for the anti-CASP4 antibody binding site. Since the number of sites is limited, as more sites are occupied by CASP4 from the sample, fewer sites are left to bind CASP4-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CASP4 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a protein that is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes composed of a prodomain and a large and small protease subunit. Activation of caspases requires proteolytic processing at conserved internal aspartic residues to generate a heterodimeric enzyme consisting of the large and small subunits. This caspase is able to cleave and activate its own precursor protein, as well as caspase 1 precursor. When overexpressed, this gene induces cell apoptosis. Alternative splicing results in transcript variants encoding distinct isoforms. [provided by RefSeq, Jul 2008]
