Canine Myocyte-specific enhancer factor 2C (MEF2C) ELISA Kit
Principle of the Assay: MEF2C ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-MEF2C antibody and an MEF2C-HRP conjugate. The assay sample and buffer are incubated together with MEF2C-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MEF2C concentration since MEF2C from samples and MEF2C-HRP conjugate compete for the anti-MEF2C antibody binding site. Since the number of sites is limited, as more sites are occupied by MEF2C from the sample, fewer sites are left to bind MEF2C-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MEF2C concentration in each sample is interpolated from this standard curve.

This locus encodes a member of the MADS box transcription enhancer factor 2 (MEF2) family of proteins, which play a role in myogenesis. The encoded protein, MEF2 polypeptide C, has both trans-activating and DNA binding activities. This protein may play a role in maintaining the differentiated state of muscle cells. Mutations and deletions at this locus have been associated with severe mental retardation, stereotypic movements, epilepsy, and cerebral malformation.
