(HTRF assay for recombinant BRDT (21-137) protein, GST-Tag activity 1 uM H4K5/8/12/16(ac4) peptide was incubated with different concentrations of BRDT (21-137) protein, GST-tag in a 10 ul reaction system containing 50 mM HEPES-NaOH pH 7.4, 0.1% BSA for 1 hour, then 10 ul GST antibody and SA-XL665 mixture (each 1:100 dilution in the reaction buffer) was added to each reaction system and incubated for 30 min. All the operations and reactions were performed at room temperature. HTRF assay was used for detection.)

Background: BRDT is similar to the RING3 protein family. It possesses two bromodomain motifs and a PEST sequence (a cluster of proline, glutamic acid, serine, and threonine residues), characteristic of proteins that undergo rapid intracellular degradation. The bromodomain functions as a 'reader' of epigenetic histone marks and regulates chromatin structure and gene expression by linking associated proteins to the recognized acetylated nucleosomal targets. It shows binding specificity for acetylated H2BK12/15, H3K9/14/18/23, H4K12, H4K5/8, H4K8/12, H4K12/16, H4K5/12, H4K5/8/12/16 and H4K5/8/12/16.

NCBI Description

BRDT is similar to the RING3 protein family. It possesses 2 bromodomain motifs and a PEST sequence (a cluster of proline, glutamic acid, serine, and threonine residues), characteristic of proteins that undergo rapid intracellular degradation. The bromodomain is found in proteins that regulate transcription. Several transcript variants encoding multiple isoforms have been found for this gene. [provided by RefSeq, Jun 2011]

Testis-specific chromatin protein that specifically binds histone H4 acetylated at 'Lys-5' and 'Lys-8' (H4K5ac and H4K8ac, respectively) and plays a key role in spermatogenesis (PubMed:22464331, PubMed:22901802). Required in late pachytene spermatocytes: plays a role in meiotic and post-meiotic cells by binding to acetylated histones at the promoter of specific meiotic and post-meiotic genes, facilitating their activation at the appropriate time (PubMed:22901802). In the post-meiotic phase of spermatogenesis, binds to hyperacetylated histones and participates in their general removal from DNA (PubMed:22901802). Also recognizes and binds a subset of butyrylated histones: able to bind histone H4 butyrylated at 'Lys-8' (H4K8ac), while it is not able to bind H4 butyrylated at 'Lys-5' (H4K5ac) (). Also acts as a component of the splicing machinery in pachytene spermatocytes and round spermatids and participates in 3'-UTR truncation of specific mRNAs in post-meiotic spermatids (). Required for chromocenter organization, a structure comprised of peri-centromeric heterochromatin.
