Intended Uses: This RANK ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine RANK. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: RANK ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-RANK antibody and an RANK-HRP conjugate. The assay sample and buffer are incubated together with RANK-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the RANK concentration since RANK from samples and RANK-HRP conjugate compete for the anti-RANK antibody binding site. Since the number of sites is limited, as more sites are occupied by RANK from the sample, fewer sites are left to bind RANK-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The RANK concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the TNF-receptor superfamily. This receptors can interact with various TRAF family proteins, through which this receptor induces the activation of NF-kappa B and MAPK8/JNK. This receptor and its ligand are important regulators of the interaction between T cells and dendritic cells. This receptor is also an essential mediator for osteoclast and lymph node development. Mutations at this locus have been associated with familial expansile osteolysis, autosomal recessive osteopetrosis, and Paget disease of bone. Alternatively spliced transcript variants have been described for this locus. [provided by RefSeq, Aug 2012]
