This assay has high sensitivity and excellent specificity for detection of CD81. No significant cross-reactivity or interference between CD81 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between CD81 and all the analogues, therefore, cross reaction may still exist in some cases.

Intended Uses: This CD81 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Bovine CD81. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: CD81 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-CD81 antibody and an CD81-HRP conjugate. The assay sample and buffer are incubated together with CD81-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CD81 concentration since CD81 from samples and CD81-HRP conjugate compete for the anti-CD81 antibody binding site. Since the number of sites is limited, as more sites are occupied by CD81 from the sample, fewer sites are left to bind CD81-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CD81 concentration in each sample is interpolated from this standard curve.

CD81: May play an important role in the regulation of lymphoma cell growth. Interacts with a 16-kDa Leu-13 protein to form a complex possibly involved in signal transduction. May act as the viral receptor for HCV. Defects in CD81 are the cause of immunodeficiency common variable type 6 (CVID6); also called antibody deficiency due to CD81 defect. CVID6 is a primary immunodeficiency characterized by antibody deficiency, hypogammaglobulinemia, recurrent bacterial infections and an inability to mount an antibody response to antigen. The defect results from a failure of B-cell differentiation and impaired secretion of immunoglobulins; the numbers of circulating B-cells is usually in the normal range, but can be low. Belongs to the tetraspanin (TM4SF) family.
