Intended Uses: This ATF3 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of RatATF3. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: ATF3 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ATF3 antibody and an ATF3-HRP conjugate. The assay sample and buffer are incubated together with ATF3-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ATF3 concentration since ATF3 from samples and ATF3-HRP conjugate compete for the anti-ATF3 antibody binding site. Since the number of sites is limited, as more sites are occupied by ATF3 from the sample, fewer sites are left to bind ATF3-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ATF3 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the mammalian activation transcription factor/cAMP responsive element-binding (CREB) protein family of transcription factors. This gene is induced by a variety of signals, including many of those encountered by cancer cells, and is involved in the complex process of cellular stress response. Multiple transcript variants encoding different isoforms have been found for this gene. It is possible that alternative splicing of this gene may be physiologically important in the regulation of target genes. [provided by RefSeq, Apr 2011]

Isoform 2 activates transcription presumably by sequestering inhibitory cofactors away from the promoters. Belongs to the bZIP family. ATF subfamily. 4 isoforms of the human protein are produced by alternative splicing.
