Intended Uses: For the quantitative detection of Human Arginase-1 (ARG1) concentration in serum, plasma and other biological fluids.

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitate ARG1 in Human serum, plasma. An antibody specific for ARG1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ARG1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ARG1 is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ARG1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

Arginase catalyzes the hydrolysis of arginine to ornithine and urea. At least two isoforms of mammalian arginase exist (types I and II) which differ in their tissue distribution, subcellular localization, immunologic crossreactivity and physiologic function. The type I isoform encoded by this gene, is a cytosolic enzyme and expressed predominantly in the liver as a component of the urea cycle. Inherited deficiency of this enzyme results in argininemia, an autosomal recessive disorder characterized by hyperammonemia. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Sep 2011]
