Principle of the Assay: KAF/AR ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-KAF/AR antibody and an KAF/AR-HRP conjugate. The assay sample and buffer are incubated together with KAF/AR-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the KAF/AR concentration since KAF/AR from samples and KAF/AR-HRP conjugate compete for the anti-KAF/AR antibody binding site. Since the number of sites is limited, as more sites are occupied by KAF/AR from the sample, fewer sites are left to bind KAF/AR-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The KAF/AR concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the epidermal growth factor family. It is an autocrine growth factor as well as a mitogen for astrocytes, Schwann cells and fibroblasts. It is related to epidermal growth factor (EGF) and transforming growth factor alpha (TGF-alpha). The protein interacts with the EGF/TGF-alpha receptor to promote the growth of normal epithelial cells, and it inhibits the growth of certain aggressive carcinoma cell lines. It also functions in mammary gland, oocyte and bone tissue development. This gene is associated with a psoriasis-like skin phenotype, and is also associated with other pathological disorders, including various types of cancers and inflammatory conditions.
