Principle of the Assay: ANG2 ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-ANG2 antibody and an ANG2-HRP conjugate. The assay sample and buffer are incubated together with ANG2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ANG2 concentration since ANG2 from samples and ANG2-HRP conjugate compete for the anti-ANG2 antibody binding site. Since the number of sites is limited, as more sites are occupied by ANG2 from the sample, fewer sites are left to bind ANG2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ANG2 concentration in each sample is interpolated from this standard curve.

The encoded protein disrupts the vascular remodeling ability of ANGPT1 and may induce endothelial cell apoptosis. ANGPT2: Binds to TEK/TIE2, competing for the ANGPT1 binding site, and modulating ANGPT1 signaling. Can induce tyrosine phosphorylation of TEK/TIE2 in the absence of ANGPT1. In the absence of angiogenic inducers, such as VEGF, ANGPT2-mediated loosening of cell-matrix contacts may induce endothelial cell apoptosis with consequent vascular regression. In concert with VEGF, it may facilitate endothelial cell migration and proliferation, thus serving as a permissive angiogenic signal. Interacts with TEK/TIE2, competing for the same binding site as ANGPT1. 3 isoforms of the human protein are produced by alternative splicing.
