Rat alpha Synuclein oligomer ELISA Kit
Principle of the Assay: SNCOalpha ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-SNCOalpha antibody and an SNCOalpha-HRP conjugate. The assay sample and buffer are incubated together with SNCOalpha-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SNCOalpha concentration since SNCOalpha from samples and SNCOalpha-HRP conjugate compete for the anti-SNCOalpha antibody binding site. Since the number of sites is limited, as more sites are occupied by SNCOalpha from the sample, fewer sites are left to bind SNCOalpha-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SNCOalpha concentration in each sample is interpolated from this standard curve.

Alpha-synuclein is a member of the synuclein family, which also includes beta- and gamma-synuclein. Synucleins are abundantly expressed in the brain and alpha- and beta-synuclein inhibit phospholipase D2 selectively. SNCA may serve to integrate presynaptic signaling and membrane trafficking. Defects in SNCA have been implicated in the pathogenesis of Parkinson disease. SNCA peptides are a major component of amyloid plaques in the brains of patients with Alzheimer's disease. Four alternatively spliced transcripts encoding two different isoforms have been identified for this gene. [provided by RefSeq, Mar 2009]
