Principle of the Assay: SNCa ELISA kit applies the competitive enzyme immunoassay technique utilizing a monoclonal anti-SNCa antibody and an SNCa-HRP conjugate. The assay sample and buffer are incubated together with SNCa-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the SNCa concentration since SNCa from samples and SNCa-HRP conjugate compete for the anti-SNCa antibody binding site. Since the number of sites is limited, as more sites are occupied by SNCa from the sample, fewer sites are left to bind SNCa-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The SNCa concentration in each sample is interpolated from this standard curve.

NCBI Description

Alpha-synuclein is a member of the synuclein family, which also includes beta- and gamma-synuclein. Synucleins are abundantly expressed in the brain and alpha- and beta-synuclein inhibit phospholipase D2 selectively. SNCA may serve to integrate presynaptic signaling and membrane trafficking. Defects in SNCA have been implicated in the pathogenesis of Parkinson disease. SNCA peptides are a major component of amyloid plaques in the brains of patients with Alzheimer's disease. Four alternatively spliced transcripts encoding two different isoforms have been identified for this gene. [provided by RefSeq, Mar 2009]
