Recombinant human ALK7 (135-end) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag.

Scientific Background: ALK7 is a type I receptor for the TGFB family of signaling molecules in which type I receptors phosphorylate cytoplasmic SMAD transcription factors, which then translocate to the nucleus and interact directly with DNA or in complex with other transcription factors (1). Alk7 was expressed in all insulin, glucagon, and somatostatin -positive cells of the pancreas (2). GDF3 regulates adipose tissue homeostasis and energy balance under nutrient overload, in part, by signaling through ALK7 and Alk7 showed reduced fat accumulation and partial resistance to diet-induced obesity, similar to Gdf3.

NCBI Description

ACVR1C is a type I receptor for the TGFB (see MIM 190180) family of signaling molecules. Upon ligand binding, type I receptors phosphorylate cytoplasmic SMAD transcription factors, which then translocate to the nucleus and interact directly with DNA or in complex with other transcription factors (Bondestam et al., 2001 [PubMed 12063393]).[supplied by OMIM, Mar 2008]

Uniprot Description

Function: Serine/threonine protein kinase which forms a receptor complex on ligand binding. The receptor complex consisting of 2 type II and 2 type I transmembrane serine/threonine kinases. Type II receptors phosphorylate and activate type I receptors which autophosphorylate, then bind and activate SMAD transcriptional regulators, SMAD2 and SMAD3. Receptor for activin AB, activin B and NODAL. Plays a role in cell differentiation, growth arrest and apoptosis. Ref.1 Ref.5

Catalytic activity: ATP + [receptor-protein] = ADP + [receptor-protein] phosphate.

Cofactor: Magnesium or manganese.

Subunit structure: Binds the type 2 receptor protein ACVR2A

By similarity.

Subcellular location: Membrane; Single-pass type I membrane protein Ref.2.

Tissue specificity: Present in pancreas, heart, colon, small intestine, ovary and the hippocampus, medulla oblongata and putamen of the brain. Isoform 1, isoform 2, isoform 3 and isoform 4 are all expressed in the placenta throughout pregnancy. Ref.1 Ref.2

Sequence similarities: Belongs to the protein kinase superfamily. TKL Ser/Thr protein kinase family. TGFB receptor subfamily.Contains 1 GS domain.Contains 1 protein kinase domain.

Product Notes

The ALK7 acvr1c (Catalog #AAA515152) is a Recombinant Protein produced from Sf9 insect cells and is intended for research purposes only. The product is available for immediate purchase. The tag for this protein is GST tag!!Expression System||Sf9 insect cells using baculovirus!!Source Note||Recombinant human ALK7 (135-end) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. AAA Biotech's ALK7 can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB). Researchers should empirically determine the suitability of the ALK7 acvr1c for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "ALK7, Recombinant Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
