Fibroblast Growth Factor, Acidic Active Protein | FGF1 active protein

Fibroblast Growth Factor, Acidic, Recombinant, Human (FGFa)

Gene Names
FGF1; ECGF; FGF-1
Purity
Highly Purified
~95% (SDS-PAGE, HPLC)
Synonyms
Fibroblast Growth Factor; Acidic; Recombinant; Human (FGFa); FGF1 active protein
Ordering
For Research Use Only!
Purity/Purification
Highly Purified
~95% (SDS-PAGE, HPLC)
Form/Format
Supplied as a lyophilized powder in PBS, pH 7.4. Reconstitute with 5mM Tris, pH 7.4, 0.1% BSA to 0.25-0.5 mg/ml.
Activity
ED50 <10 ng/ml (> 1 x 105 units/mg). Biological activity is determined by measuring the FGF-a dose dependent stimulation of 3H-thymidine incorporation into BaF3 cell expressing FGF receptors. For most in vitro applications, a concentration range of 1.0-50 ng/ml is recommended. FGF-a is a heparin-binding protein which stimulates the proliferation of mesenchymal cells, neuroectodermal cells and endothelial cells.
Endotoxin Level
0.1ng/ug
Preparation and Storage
Lyophilized powder may be stored at -20 degree C. Stable for 12 months at -20 degree C. Reconstitute with Tris, pH 7.4, BSA. Aliquot to avoid repeated freezing and thawing. Store at -20 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Uniprot Description

Function: Plays an important role in the regulation of cell survival, cell division, angiogenesis, cell differentiation and cell migration. Functions as potent mitogen in vitro. Ref.11 Ref.12

Subunit structure: Monomer. Homodimer. Interacts with FGFR1, FGFR2, FGFR3 and FGFR4. Affinity between fibroblast growth factors (FGFs) and their receptors is increased by heparan sulfate glycosaminoglycans that function as coreceptors. Found in a complex with FGFBP1, FGF1 and FGF2. Interacts with FGFBP1. Part of a Cu2+-dependent multiprotein aggregate containing FGF1, S100A13 and SYT1. Interacts with S100A13. Interacts with FGFBP1

By similarity. Interacts with LRRC59

By similarity. Interacts with CSNKA, CSNKB and FIBP

By similarity. While binding with LRRC59, CSNKA and FIBP seem mutually exclusive, CSNKB and FIBP may cooperatively interact with FGF1

By similarity. Interacts with SYT1. Ref.11

Subcellular location: Secreted. Cytoplasm

By similarity. Cytoplasm › cell cortex

By similarity. Cytoplasm › cytosol. Nucleus. Note: Lacks a cleavable signal sequence. Within the cytoplasm, it is transported to the cell membrane and then secreted by a non-classical pathway that requires Cu2+ ions and S100A13. Secreted in a complex with SYT1. Binding of exogenous FGF1 to FGFR facilitates endocytosis followed by translocation of FGF1 across endosomal membrane into the cytosol. Nuclear import from the cytosol requires the classical nuclear import machinery, involving proteins KPNA1 and KPNB1, as well as LRRC59

By similarity. Ref.11 Ref.12

Post-translational modification: In the nucleus, phosphorylated by PKC/PRKCD

By similarity.

Sequence similarities: Belongs to the heparin-binding growth factors family.

Product Notes

The FGF1 fgf1 (Catalog #AAA650508) is an Active Protein and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "Fibroblast Growth Factor, Acidic, Active Protein" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
