Unit Definition
One unit of the recombinant caspase-4 is the enzyme activity that cleaves 1 nmol of the caspase substrate WEHD-pNA (pNA: pnitroanaline) per hour at 37 degree C in a reaction solution containing 50mM Hepes, pH 7.2, 50mM sodium chloride, 0.1% CHAPS, 10mM EDTA, 5% Glycerol, 10mM DTT.
Preparation and Storage
Lyophilized powder may be stored at 4 degree C for short-term only. For long term storage, store at -70 degree C for 1 year. To avoid multiple freeze thaw cycles, reconstituted product should be aliquoted and stored at 70 degree C. Aliquots are stable for 12 months at -70 degree C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Caspase-4 (also know as ICErelII, TX, Ich-2) is the member of the caspase family of cysteine proteases. Caspase-4 exists in cells as an inactive pro-enzyme. The pro-enzyme is matured by proteolysis to yield large and small subunits. The active enzyme is a heterotetramer consisting of two large and two small subunits. To date the mechanism of caspase-4 activation and roles of caspase-4 in apoptosis are poorly understood. Active recombinant human caspase-4 was expressed in E. coli. The expressed caspase-4 spontaneously undergoes auto-processing to yield subunits caracteristic of the native enzyme. The active caspase-4 is routinely tested at Untied States Biological for its ability to enzymatically cleaves the substrate WEHD-AFC or WEHD-pNA.

NCBI Description

This gene encodes a protein which is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein cleaves and activates caspases 3 and 7, and the protein itself is processed by caspase 8. Mutations in this gene are associated with type IIA autoimmune lymphoproliferative syndrome, non-Hodgkin lymphoma and gastric cancer. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. [provided by RefSeq, Apr 2011]

May participate in the granzyme B apoptotic pathways. Cleaves and activates caspase- 3, -4, -6, -7, -8, and -9. Hydrolyzes the small- molecule substrates, Tyr-Val-Ala-Asp-|-AMC and Asp-Glu-Val-Asp-|-AMC. Heterotetramer that consists of two anti-parallel arranged heterodimers, each one formed by a 23/17 kDa (p23/17) (depending on the splicing events) and a 12 kDa (p12) subunit. Self-associates. Interacts with FADD and CASP8. Found in a Fas signaling complex consisting of FAS, FADD, CASP8 and CASP10. Detectable in most tissues. Lowest expression is seen in brain, kidney, prostate, testis and colon. Belongs to the peptidase C14A family. 6 isoforms of the human protein are produced by alternative splicing.
