Intended Uses: This ACACalpha ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Mouse ACACalpha. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: ACACalpha ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-ACACalpha antibody and an ACACalpha-HRP conjugate. The assay sample and buffer are incubated together with ACACalpha-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the ACACalpha concentration since ACACalpha from samples and ACACalpha-HRP conjugate compete for the anti-ACACalpha antibody binding site. Since the number of sites is limited, as more sites are occupied by ACACalpha from the sample, fewer sites are left to bind ACACalpha-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The ACACalpha concentration in each sample is interpolated from this standard curve.

NCBI Description

Acetyl-CoA carboxylase (ACC) is a complex multifunctional enzyme system. ACC is a biotin-containing enzyme which catalyzes the carboxylation of acetyl-CoA to malonyl-CoA, the rate-limiting step in fatty acid synthesis. There are two ACC forms, alpha and beta, encoded by two different genes. ACC-alpha is highly enriched in lipogenic tissues. The enzyme is under long term control at the transcriptional and translational levels and under short term regulation by the phosphorylation/dephosphorylation of targeted serine residues and by allosteric transformation by citrate or palmitoyl-CoA. Multiple alternatively spliced transcript variants divergent in the 5' sequence and encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

Phosphorylation by AMPK or PKA inhibits the enzymatic activity of ACC. ACC-alpha is the predominant isoform in liver, adipocyte and mammary gland. ACC-beta is the major isoform in skeletal muscle and heart. Phosphorylation regulates its activity.
