Similarity
The PX domain is required for podosome localization because of its ability to bind phosphatidylinositol 3-phosphate (PtdIns(3)P) and phosphatidylinositol 3, 4-bisphosphate (PtdIns(3, 4)P2) and, to a lesser extent, phosphatidylinositol 4-phosphate (PtdIns(4)P), phosphatidylinositol 5-phosphate (PtdIns(5)P), and phosphatidylinositol 3, 5-bisphosphate (PtdIns(3, 5)P2). Binds to the third intramolecular SH3 domain (By similarity). Belongs to the SH3PXD2 family.
Post Translational Modifications
Phosphorylated in SRC-transformed cells.
Subunit Structure
Interacts with ADAM15 (By similarity). Interacts with NOXO1. Interacts (via SH3 domains) with NOXA1; the interaction is direct. Interacts with FASLG.
Tissue Specificity
Expressed in fibroblasts.
Function
Adapter protein involved in invadopodia and podosome formation and extracellular matrix degradation. Binds matrix metalloproteinases (ADAMs), NADPH oxidases (NOXs) and phosphoinositides. Acts as an organizer protein that allows NOX1- or NOX3-dependent reactive oxygen species (ROS) generation and ROS localization. Plays a role in mitotic clonal expansion during the immediate early stage of adipocyte differentiation (By similarity).
Note
Cytoplasmic in normal cells and localizes to podosomes in SRC-transformed cells.
Preparation and Storage
Store at -20 degree C. Stable for 12 months from date of receipt.

NCBI Description

This gene encodes an adapter protein that is characterized by a PX domain and four Src homology 3 domains. The encoded protein is required for podosome formation and is involved in cell adhesion and migration of numerous cell types. Mutations in this gene are the cause of Frank-ter Haar syndrome. [provided by RefSeq]

Uniprot Description

SH3PXD2B: Adapter protein involved in invadopodia and podosome formation and extracellular matrix degradation. Binds matrix metalloproteinases (ADAMs), NADPH oxidases (NOXs) and phosphoinositides. Acts as an organizer protein that allows NOX1- or NOX3-dependent reactive oxygen species (ROS) generation and ROS localization. Plays a role in mitotic clonal expansion during the immediate early stage of adipocyte differentiation. Defects in SH3PXD2B are the cause of Frank-Ter Haar syndrome (FTHS). It is a syndrome characterized by brachycephaly, wide fontanels, prominent forehead, hypertelorism, prominent eyes, macrocornea with or without glaucoma, full cheeks, small chin, bowing of the long bones and flexion deformity of the fingers. Belongs to the SH3PXD2 family.

Product Notes

The SH3PXD2B sh3pxd2b (Catalog #AAA9629183) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The SH3PXD2B Antibody reacts with Human, Mouse, Rat and may cross-react with other species as described in the data sheet. AAA Biotech's SH3PXD2B can be used in a range of immunoassay formats including, but not limited to, ELISA (EIA)(peptide). ELISA(peptide): 1:20000-1:40000. Researchers should empirically determine the suitability of the SH3PXD2B sh3pxd2b for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "SH3PXD2B, Polyclonal Antibody" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
