NAMPT blocking peptide

NAMPT Antibody (Center) Blocking Peptide

Gene Names
NAMPT; VF; PBEF; PBEF1; VISFATIN; 1110035O14Rik
Synonyms
NAMPT; NAMPT Antibody (Center) Blocking Peptide; Nicotinamide phosphoribosyltransferase; NAmPRTase; Nampt; Pre-B-cell colony-enhancing factor 1; Pre-B cell-enhancing factor; Visfatin; PBEF; PBEF1; NAMPT blocking peptide
Ordering
Specificity
The synthetic peptide sequence used to generate the antibody was selected from the Center region of human NAMPT. A 10 to 100 fold molar excess to antibody is recommended. Precise conditions should be optimized for a particular assay.
Form/Format
Synthetic peptide was lyophilized with 100% acetonitrile and is supplied as a powder. Reconstitute with 0.1 ml DI water for a final concentration of 1 mg/ml.
Sequence Length
491
Cellular Location
Nucleus. Cytoplasm. Secreted. Note: Under non-inflammatory conditions, visfatin predominantly exhibits a granular pattern within the nucleus. Secreted by endothelial cells upon IL-1beta stimulation. Abundantly secreted in milk, reaching 100-fold higher concentrations compared to maternal serum
Tissue Location
Expressed in large amounts in bone marrow, liver tissue, and muscle. Also present in heart, placenta, lung, and kidney tissues
Preparation and Storage
Maintain refrigerated at 2-8 degree C for up to 6 months. For long term storage store at -20 degree C.
Related Product Information for NAMPT blocking peptide
Catalyzes the condensation of nicotinamide with 5- phosphoribosyl-1-pyrophosphate to yield nicotinamide mononucleotide, an intermediate in the biosynthesis of NAD. It is the rate limiting component in the mammalian NAD biosynthesis pathway. The secreted form behaves both as a cytokine with immunomodulating properties and an adipokine with anti-diabetic properties, it has no enzymatic activity, partly because of lack of activation by ATP, which has a low level in extracellular space and plasma. Plays a role in the modulation of circadian clock function. NAMPT-dependent oscillatory production of NAD regulates oscillation of clock target gene expression by releasing the core clock component: CLOCK-ARNTL/BMAL1 heterodimer from NAD-dependent SIRT1-mediated suppression (By similarity).

NCBI Description

This gene encodes a protein that catalyzes the condensation of nicotinamide with 5-phosphoribosyl-1-pyrophosphate to yield nicotinamide mononucleotide, one step in the biosynthesis of nicotinamide adenine dinucleotide. The protein belongs to the nicotinic acid phosphoribosyltransferase (NAPRTase) family and is thought to be involved in many important biological processes, including metabolism, stress response and aging. This gene has a pseudogene on chromosome 10. [provided by RefSeq, Feb 2011]

Uniprot Description

PBEF: an ubiquitous protein originally described as a cytokine which acts on early B-lineage precursor cells. Other reports indicate that PBEF is not a cytokine-like secreted protein but an intracellular protein associated with the cell cycle. Contains a catalytically active nicotinate phosphoribosyltransferase domain, an enzyme involved in nicotinamide adenine dinucleotide (NAD) biosynthesis.

Protein type: EC 2.4.2.12; Cell cycle regulation; Cytokine; Cofactor and Vitamin Metabolism - nicotinate and nicotinamide; Transferase

Chromosomal Location of Human Ortholog: 7q22.3

Cellular Component: cell junction; cytosol; nucleoplasm

Molecular Function: nicotinamide phosphoribosyltransferase activity; protein binding

Biological Process: cell-cell signaling; circadian regulation of gene expression; circadian rhythm; insulin receptor signaling pathway; nicotinamide metabolic process; positive regulation of cell proliferation; positive regulation of nitric-oxide synthase biosynthetic process; positive regulation of transcription from RNA polymerase II promoter; signal transduction

Product Notes

The NAMPT nampt (Catalog #AAA9227973) is a Blocking Peptide and is intended for research purposes only. The product is available for immediate purchase. It is sometimes possible for the material contained within the vial of "NAMPT, Blocking Peptide" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
