Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for KLK4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any KLK4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for KLK4 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of KLK4 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

Kallikreins are a subgroup of serine proteases having diverse physiological functions. Growing evidence suggests that many kallikreins are implicated in carcinogenesis and some have potential as novel cancer and other disease biomarkers. This gene is one of the fifteen kallikrein subfamily members located in a cluster on chromosome 19. In some tissues its expression is hormonally regulated. The expression pattern of a similar mouse protein in murine developing teeth supports a role for the protein in the degradation of enamel proteins. Alternate splice variants for this gene have been described, but their biological validity has not been determined. [provided by RefSeq, Jul 2008]

Uniprot Description

KLK4: Involved in enamel formation. Defects in KLK4 are the cause of amelogenesis imperfecta hypomaturation type 2A1 (AI2A1). AI2A1 is an autosomal recessive defect of enamel formation. The disorder involves both primary and secondary dentitions. The teeth have a shiny agar jelly appearance and the enamel is softer than normal. Brown pigment is present in middle layers of enamel. Belongs to the peptidase S1 family. Kallikrein subfamily.

Chromosomal Location of Human Ortholog: 19q13.41

Cellular Component: extracellular region

Molecular Function: serine-type peptidase activity; metal ion binding; serine-type endopeptidase activity

Biological Process: extracellular matrix disassembly; protein catabolic process; proteolysis

Product Notes

The Human KLK4 klk4 (Catalog #AAA917102) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA917102 ELISA Kit recognizes Human KLK4. It is sometimes possible for the material contained within the vial of "kallikrein-related peptidase 4, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
