Related Product Information for CHIT1 assay kit
Background/Introduction: Chitotriosidase (CHIT1 or chitinase-1), is a mammalian enzyme belonging to glycosyl hydrolase-18 family (EC 3.2.1.14), which can hydrolyze the beta-(1, 4)-linkage between the adjacent N-acetyl glucosamine residues of chitin. CHIT1 is synthesized exclusively by activated macrophages both in normal and inflammatory conditions and, as a host defense factor, CHIT1 plays a pivotal role in infectious disease such as malaria and fungi infectious. Increased CHIT1 activity has also been found in patients suffering diseases such as bronchial asthma and atherosclerosis. CHIT1 has been used as a valuable test biomarker for monitoring the therapeutic efficacy of treatments for Gaucher's disease or beta-glucocerebrosidase deficiency. Chitotriosidase Activity Assay, utilizes a fluorogenic substrate that can be hydrolyzed by chitinase and a set of proprietary assay buffers that can distinguish specific CHIT1 activity from other hydrolases including Acidic Mammalian Chitinase. The kit provides a simple, specific, sensitive assay that can detect as low as 0.2 mU/ml of CHIT1 in a variety of biological samples.

NCBI Description

Chitotriosidase is secreted by activated human macrophages and is markedly elevated in plasma of Gaucher disease patients. The expression of chitotriosidase occurs only at a late stage of differentiation of monocytes to activated macrophages in culture. Human macrophages can synthesize a functional chitotriosidase, a highly conserved enzyme with a strongly regulated expression. This enzyme may play a role in the degradation of chitin-containing pathogens. Several alternatively spliced transcript variants have been described for this gene. [provided by RefSeq, Jan 2012]

Uniprot Description

CHIT1: Degrades chitin, chitotriose and chitobiose. May participate in the defense against nematodes and other pathogens. Isoform 3 has no enzymatic activity. Belongs to the glycosyl hydrolase 18 family. Chitinase class II subfamily. 3 isoforms of the human protein are produced by alternative splicing.

Product Notes

The Mammalian CHIT1 chit1 (Catalog #AAA846798) is an Assay Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA846798 Assay Kit recognizes Mammalian CHIT1. Measurement of Chitotriosidase activity in various biological samples and purified/crude enzyme preparations. Researchers should empirically determine the suitability of the CHIT1 chit1 for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process. It is sometimes possible for the material contained within the vial of "Chitotriosidase, Assay Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
