Principle of the Assay||This kit was based on sandwich enzyme-linked immune-sorbent assay technology. anti-Human BANP antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-Human BANP antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human BANP amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of Human BANP can be calculated.

The protein forms a complex with p53 and negatively regulates p53 transcription, and functions as a tumor suppressor and cell cycle regulator. Represses cyclin D1 transcription by recruiting HDAC1 to its promoter, thereby diminishing H3K9ac, H3S10ph and H4K8ac levels. Promotes TP53 'Ser-15' phosphorylation and nuclear accumulation, which causes cell cycle arrest. Belongs to the BANP/SMAR1 family. 6 isoforms of the human protein are produced by alternative splicing.
