Bovine Microtubule-associated protein 1B ELISA Kit
Principle of the Assay: MAP1B ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-MAP1B antibody and an MAP1B-HRP conjugate. The assay sample and buffer are incubated together with MAP1B-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the MAP1B concentration since MAP1B from samples and MAP1B-HRP conjugate compete for the anti-MAP1B antibody binding site. Since the number of sites is limited, as more sites are occupied by MAP1B from the sample, fewer sites are left to bind MAP1B-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The MAP1B concentration in each sample is interpolated from this standard curve.

This gene encodes a protein that belongs to the microtubule-associated protein family. The proteins of this family are thought to be involved in microtubule assembly, which is an essential step in neurogenesis. The product of this gene is a precursor polypeptide that presumably undergoes proteolytic processing to generate the final MAP1B heavy chain and LC1 light chain. Gene knockout studies of the mouse microtubule-associated protein 1B gene suggested an important role in development and function of the nervous system. [provided by RefSeq, Jul 2008]

MAP1B: the function of brain MAPS is essentially unknown. Phosphorylated MAP1B may play a role in the cytoskeletal changes that accompany neurite extension. Possibly MAP1B Binds to at least two tubulin subunits in the polymer, and this bridging of subunits might be involved in nucleating microtubule polymerization and in stabilizing microtubules.
