Reactivity
Canine
Specificity
This assay has high sensitivity and excellent specificity for detection of BMP-10. No significant cross-reactivity or interference between BMP-10 and analogues was observed. NOTE: Limited by current skills and knowledge, it is impossible for us to complete the cross-reactivity detection between BMP-10 and all the analogues, therefore, cross reaction may still exist in some cases.

Serum, plasma, cell culture supernatants, body fluid and tissue homogenate
Assay Type
Quantitative Competitive
Sensitivity
0.1 ng/mL
Preparation and Storage
Store all reagents at 2-8 degree C.

Intended Uses: This BMP-10 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Canine BMP-10. This ELISA kit for research use only, not for therapeutic or test applications!

Principle of the Assay: BMP-10 ELISA kit applies the competitive enzyme immunoassay technique utilizing a polyclonal anti-BMP-10 antibody and an BMP-10-HRP conjugate. The assay sample and buffer are incubated together with BMP-10-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the BMP-10 concentration since BMP-10 from samples and BMP-10-HRP conjugate compete for the anti-BMP-10 antibody binding site. Since the number of sites is limited, as more sites are occupied by BMP-10 from the sample, fewer sites are left to bind BMP-10-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The BMP-10 concentration in each sample is interpolated from this standard curve.

NCBI Description

The protein encoded by this gene is a member of the TGF-beta family of growth factors. Data suggest that the similar protein in mouse plays an important role in trabeculation of the embryonic heart. In human, this protein may signal through receptor serine/threonine kinases. [provided by RefSeq, Jul 2008]

Uniprot Description

BMP10: Required for maintaining the proliferative activity of embryonic cardiomyocytes by preventing premature activation of the negative cell cycle regulator CDKN1C/p57KIP and maintaining the required expression levels of cardiogenic factors such as MEF2C and NKX2-5. Acts as a ligand for ACVRL1/ALK1, BMPR1A/ALK3 and BMPR1B/ALK6, leading to activation of SMAD1, SMAD5 and SMAD8 transcription factors. Inhibits endothelial cell migration and growth. Belongs to the TGF-beta family.

Product Notes

The Canine BMP10 bmp10 (Catalog #AAA742370) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA742370 ELISA Kit recognizes Canine BMP10. It is sometimes possible for the material contained within the vial of "Bone Morphogenetic Protein 10, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
