Human C type lectin domain family 14 member A (CLEC14A) ELISA Kit
Principle of the Assay: CLDN5 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-CLDN5 antibody and an CLDN5-HRP conjugate. The assay sample and buffer are incubated together with CLDN5-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the CLDN5 concentration since CLDN5 from samples and CLDN5-HRP conjugate compete for the anti-CLDN5 antibody binding site. Since the number of sites is limited, as more sites are occupied by CLDN5 from the sample, fewer sites are left to bind CLDN5-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The CLDN5 concentration in each sample is interpolated from this standard curve.

Members of this family share a common protein fold and have diverse functions, such as cell adhesion, cell-cell signalling, glycoprotein turnover, and roles in inflammation and immune response. This family member plays a role in cell-cell adhesion and angiogenesis. It functions in filopodia formation, cell migration and tube formation. Due to its presence at higher levels in tumor endothelium than in normal tissue endothelium, it is considered to be a candidate for tumor vascular targeting. [provided by RefSeq, Jan 2012]
