Intended Uses: This FBXL2 ELISA kit is a 1.5 hour solid-phase ELISA designed for the quantitative determination of Human FBXL2. This ELISA kit for research use only!

Principle of the Assay: FBXL2 ELISA kit applies the competitive enzyme immunoassay technique utilizing an anti-FBXL2 antibody and an FBXL2-HRP conjugate. The assay sample and buffer are incubated together with FBXL2-HRP conjugate in pre-coated plate for one hour. After the incubation period, the wells are decanted and washed five times. The wells are then incubated with a substrate for HRP enzyme. The product of the enzyme-substrate reaction forms a blue colored complex. Finally, a stop solution is added to stop the reaction, which will then turn the solution yellow. The intensity of color is measured spectrophotometrically at 450nm in a microplate reader. The intensity of the color is inversely proportional to the FBXL2 concentration since FBXL2 from samples and FBXL2-HRP conjugate compete for the anti-FBXL2 antibody binding site. Since the number of sites is limited, as more sites are occupied by FBXL2 from the sample, fewer sites are left to bind FBXL2-HRP conjugate. A standard curve is plotted relating the intensity of the color (O.D.) to the concentration of standards. The FBXL2 concentration in each sample is interpolated from this standard curve.

NCBI Description

This gene encodes a member of the F-box protein family which is characterized by an approximately 40 amino acid motif, the F-box. The F-box proteins constitute one of the four subunits of ubiquitin protein ligase complex called SCFs (SKP1-cullin-F-box), which function in phosphorylation-dependent ubiquitination. The F-box proteins are divided into 3 classes: Fbws containing WD-40 domains, Fbls containing leucine-rich repeats, and Fbxs containing either different protein-protein interaction modules or no recognizable motifs. The protein encoded by this gene belongs to the Fbls class and, in addition to an F-box, contains 12 tandem leucine-rich repeats. Alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jan 2010]

FBXL2: Calcium-activated substrate recognition component of a SCF (SKP1-cullin-F-box protein) E3 ubiquitin-protein ligase complex which mediates the ubiquitination and subsequent proteasomal degradation of target proteins. Unlike many F-box proteins, FBXL2 doesn't seem to target phosphodegron within its substrates but rather calmodulin-binding motifs. Targets PCYT1A for its monoubiquitination and degradation, this is antagonized by calmodulin. Targets the cyclins CCND2 and CCND3 for polyubiquitination and degradation, leading to cell-cycle arrest in G(0), also antagonized by calmodulin. Binds to hepatitis C virus non-structural protein 5A (NS5A) in a reaction crucial for hepatitis C virus RNA replication.
