Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MAGE-3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MAGE-3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MAGE-3 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAGE-3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

NCBI Description

This gene is a member of the MAGEA gene family. The members of this family encode proteins with 50 to 80% sequence identity to each other. The promoters and first exons of the MAGEA genes show considerable variability, suggesting that the existence of this gene family enables the same function to be expressed under different transcriptional controls. The MAGEA genes are clustered at chromosomal location Xq28. They have been implicated in some hereditary disorders, such as dyskeratosis congenita. [provided by RefSeq, Jul 2008]

Uniprot Description

Function: Proposed to enhance ubiquitin ligase activity of RING-type zinc finger-containing E3 ubiquitin-protein ligases. May enhance ubiquitin ligase activity of TRIM28 and stimulate p53/TP53 ubiquitination by TRIM28. Proposed to act through recruitment and/or stabilization of the Ubl-conjugating enzyme (E2) at the E3:substrate complex. May play a role in embryonal development and tumor transformation or aspects of tumor progression. In vitro promotes cell viability in melanoma cell lines. Antigen recognized on a melanoma by autologous cytolytic T-lymphocytes.1 PublicationManual assertion based on experiment in:Ref.9

Product Notes

The Human MAGEA3 magea3 (Catalog #AAA705585) is an ELISA Kit and is intended for research purposes only. The product is available for immediate purchase. The AAA705585 ELISA Kit recognizes Human MAGEA3. It is sometimes possible for the material contained within the vial of "melanoma antigen family A, 3, ELISA Kit" to become dispersed throughout the inside of the vial, particularly around the seal of said vial, during shipment and storage. We always suggest centrifuging these vials to consolidate all of the liquid away from the lid and to the bottom of the vial prior to opening. Please be advised that certain products may require dry ice for shipping and that, if this is the case, an additional dry ice fee may also be required.
